Alpaca a punch for humans against COVIDGRANT McARTHURALPACAS may be a secret weapon in the fight against COVID-19, as scientists try to adapt their unique immune system into a nasal spray to protect humans.Having identified how alpacas are able to fight the SARS-CoV-2 virus, a Melbourne collaboration has now been able to harness the properties of their immune system to successfully shield mice from COVID-19.Work led by the Walter and Eliza Hall Institute is now moving to adapt the breakthrough to humans to protect against current and more infectious variants of COVID- 19 now emerging.Unlike humans and most other animals, alpacas release millions of tiny immune proteins known as nanobodies to fight off infections.As well as being much smaller than human antibodies, the alpaca nanobodies are also much more stable, which WEHI Associate Professor Wai-Hong Tham said made them better suited to attaching to the spikes of coronavirus so it cannot infect cells.β€œAlpacas make the tiniest antibodies that currently exist,” Assoc Prof Tham said.β€œThey are really small and they have different ways they can actually bind to spike proteins to stop coronavirus.β€œIf I deliver the nanobodies a day before infection, what we see in the preclinical model is we have basically no viral load. And, because they are a bit smaller, they have a property of being very stable and also cheaper to manufacture.”While the exact reasons why are unknown, alpacas, camels and other members of the camelid family naturally produce nanobodies which are smaller fragments of the same antibodies produced by humans. After immunising a herd of Gippsland alpacas with a synthetic version of the SARSCoV-2 virus – which included its notorious spike protein but not the inner workings needed to grow – the WEHI scientists were able to map which nanobodies the animals created to fight off the invader.With the help of the Australian Synchrotron, scientists examined about one million different nanobodies produced by the infected alpacas, identifying 50 that were most active in overcoming coronavirus.Two of those nanobodies specifically targeted the spike protein, which the scientists focused on. ...
